Dr. Wang is an associate professor in the Department of Internal Medicine and the Michigan Nanotechnology Institute for Medicine and Biological Sciences. She has morethan 20 years of research and teaching experience in autoimmune thyroid diseaseand thyroid cancer. In the last ten years, she has actively extended herresearch to include functionalized nanoparticles as a diagnostic andtherapeutic approach for cancer cell targeting and imaging. Dr. Wang has beenthe principal investigator of a project funded by the Department of Defensethat utilizes nanoemulsion as a wound and burn decontamination agent. She isalso as a co-investigator for the nanoemulsion adjuvant development programfunded by BAA-NIAID.

Many breast and lung cancersoverexpress human epidermal growth factor receptor-2 (HER-2). As such, in thisstudy we sought to enhance and specifically design early detection imagingagents against these cancers by modulating the humanized anti-HER-2 antibody Herceptin with gadolinium gold dendrimer entrapped nanoparticles (Gd-AuDENP).Able to enhance computerizedtomography and magnetic resonance contrast for detecting cancers early, Gd-AuDENPs were synthesized by entrapping gold nanoparticles (AuNPs) in G5 PAMAMdendrimers using sodium borohydride reduction chemistry while gadolinium waschelated into DOTA conjugated to the dendrimer. Herceptin was modified with a5-azidopentanoic acid linker and linked to the G5 PAMAM dendrimer via copperclick chemistry. Treatment with Gd-Au DENP-Herceptin in vitro demonstrated the conjugate specifically and exclusively binds to cell lines overexpressingHER-2. Subsequent internalization occurred, whereas untargeted Gd-Au DENPsneither bound to nor were internalized into cells lines overexpressing HER-2.These data therefore show a facile method for conjugation of Herceptin to adendrimer based imaging agent with capacity for versatile modification viaclick chemistry. This conjugate could serve in the future simultaneously as adrug delivery agent and locator especially of metastatic and early stagecancers.
